大豆肽在不同金属离子螯合亲和层析介质(Cu2+、Fe3+、Zn2+和Ca2+)吸附能力的比较

摘要:以琼脂糖凝胶为固相支持物,亚氨基二乙酸为螯合剂,用Cu2+、Fe3+、Zn2+和Ca2+ 4种金属离子制成琼脂凝胶金属离子亲和层析柱,并对层析柱上螯合的金属离子量以及这4种金属离子亲和层析柱对大豆肽组分的吸附效果和分离效果进行了比较。结果发现,金属离子的螯合量及固定金属离子亲和层析柱对大豆肽组分的亲和效果均为Fe3+>Cu2+>Zn2+>Ca2+。

Effect of calcium, iron, zinc, copper on the adsorption of soybean protein hydrolysates on immobilized metal affinity chromatography
Abstract:4 kinds of metal ions were used in immobilized metal ion affinity chromatography sorbents prepared with Sepharose as substrate. Then the amounts of immobilized metal ion on IDA-Sepharose and binding amounts of soybean peptide to immobilized IDM-Mn+ adsorbents were compared. The results show that the binding ability of soybean protein peptide on the chromatography column (strong to weak) should be Fe3+>Cu2+>Zn2+>Ca2+.
