琐琐葡萄中多糖、总黄酮和总三萜的一步提取工艺与含量测定

摘要:目的:从琐琐葡萄中一步提取分离多糖(VTP)、总黄酮(VTF)和总三萜(VTT),并进行含量测定。方法:应用植物化学的方法提取分离VTP、VTF和VTT,紫外分光光度法测定多糖和总黄酮含量。结果:建立了综合应用水提醇沉法、醇提法、有机溶剂萃取法及大孔吸附树脂富集、乙醇-水梯度洗脱等方法一步提取分离出VTP、VTF和VTT的工艺;VTP粗提物中多糖含量为20.68%,RSD为1.93%,纯化物中多糖含量为36.11%,RSD为1.29%;VTF中总黄酮含量为35.28%,RSD为1.42%。结论:本研究所建立的琐琐葡萄多糖、总黄酮和总三萜的一步提取工艺简便易行、经济高效、含量较高。

The one-step extraction of polysaccharides, total flavones and total triterpene from Vitis Viniferal L and their content determination
Abstract:Objectives: To extract polysaccharides, total flavones and total triterpene from one portion of Vitis Viniferal L material and study the content of polysaccharides and total flavones. Methods: We employed phytochemistry approach to extract polysaccharides, total flavones and total triterpene from Vitis Viniferal L; Ultraviolet spectrophotometry was used to determinate the content of polysaccharides in VTP and total flavones in VTF. Results: The one-step extraction approach of VTP, VTF and VTT was set up, applying the methods of water extraction, ethanol subsidy, macroporous adsorptive resins gathering, ethanol-water elution and so on; The content of polysaccharides in VTP crude extractive and purified extractive are 20.68% and 36.11% respectively, the content of total flavones in VTF is 35.28%,RSD is 1.42%. Conclusions: The one-step extraction approach of VTP,VTF and VTT from Vitis Viniferal L established is convenient, practical, and of low cost, high efficiency and high content.
