眼用促渗透剂刺激性基于鸡胚模型的评价研究

摘要:目的:研究用鸡胚尿囊绒膜-苔盼蓝法评价眼部促渗透剂刺激性的可行性。方法:用该法对几种常用促渗透剂的刺激性进行检测,用紫外可见分光光度计在595 nm处测定苔盼蓝的吸光度。结果:冰片,吐温-80,苯扎氯铵,二甲亚砜,丙二醇的刺激性均不同,其中1%二甲亚砜的刺激性最大。1%丙二醇的促渗透效果最好。结论:鸡胚尿囊绒膜-苔盼蓝吸收法能够客观量化的对促渗透剂的刺激性进行评价。

Irritant evaluation of ocular permeation enhancers based on chick chorioallantoic membrane trypan blue test
Abstract:Objective: To study eye irritation induced by various permeation enhancers through chick chorioallantoic membrane trypan blue test (CAM-TB). Method: The irritation of some ocular permeation enhancers was evaluated by CAM-TB. Ultraviolet and visible spectrophotometer was used for analysis and the adsorption of trypan blue was measured at 595nm. Result: The irritation of penetration enhancers was different as for borneol, Tween-80, benzalkonium chloride, DMSO, propanediol. The DMSO solution of 1% w/v had the most potent irritant effect. Solution of propanediol (1% w/v) had the strongest effect of promoting penetration. Conclusion: Chorioallantoic membrane trypan blue test could quantificationally evaluate the irritation induced by various ocular permeation enhancers.
