HSCCC法分离制备鱼藤根中的两种鱼藤酮类化合物

摘要:采用高速逆流色谱(HSCCC)法,以正己烷-乙酸乙酯-甲醇-水(体积比为7:0.25:5:3)为两相溶剂系统,上相为固定相,下相为流动相,在主机转850r/min、流速2.0mL/min、检测波长254nm条件下进行分离制备,进样量为50mg,结果分离得到两种鱼藤酮类化合物,分别为:纯度为96.60%的鱼藤酮6.4mg和纯度为97.87%的鱼藤素23.4mg.

Separation and preparation of two kinds of rotenoid from the roots of Derris by high-speed counter-current chromatography
Abstract:Using the method of high-speed countercurrent chromatography, The optimized two-phase solvent system, flow rate, revolution speed ,detected wavelengthand injection amount of HSCCC were hexane-ethylacetate-methanol-water(7:0.5:5:3, V/V),2ml/min and 850r/min and 50mg, respectively. The upper phase was used as the stationary phase, while the lower phase was the mobile phase of HSCCC.Under these conditions,The two kinds of rotenoid compounds are respectively: the purities of rotenone and deguelin were 96.60%(6.4mg)and 97.87%(23.4mg).
