细胞癌血清差异蛋白的实验研究

摘要:目的 探讨应用磁性微球、MALDI技术筛选肾细胞癌患者血清中可用于诊断的差异蛋白峰。方法:99份血清标本,其中肾细胞癌62份,肾脏良性占位37份,采用弱阳离子磁性微球捕获肾细胞癌患者血清中的蛋白,MALDI-TOF-MS质谱仪检测绘制蛋白峰,应用Biomarker Wizard 3.1和Biomarker Patterns Software 5.0分析数据,建立肾细胞癌诊断模型,盲法验证。结果:在肾细胞癌与对照组之间找到7个差异蛋白峰(P<0.05),应用质荷比为2945.35 、15340.8、6984.51、5819.23的4个蛋白峰建立肾细胞癌诊断模型,该模型区分肾细胞癌与良性占位的敏感性83.0%、特异性84.6%,双盲试验验证,该模型诊断肾细胞癌的敏感性80.0%、特异性81.8%。 结论:采用磁性微球与MALDI技术可以检测肾细胞癌血清中差异蛋白峰,并建立敏感性、特异性较高的肾细胞癌诊断模型。

Investigation the differential proteins of renal cell carcinoma
Abstract:Objective To screen for the differential proteins peaks for diagnosis of renal cell carcinoma (RCC) using technology combining magnetic beads with MALDI-TOF-MS. Mathods Technology combining magnetic beads (WCX) with MALDI-TOF-MS was used to profile and compare the serum protein from 62 patients with RCC, 37 patients with renal benign space occupying. The data were analyzed using Biomarker Wizard 3.1and Biomarker Patterns Software 5.0. Model diagnosis for RCC was constructed and evaluated. Results A total of 7 differential protein peaks were identified that were related to RCC (P<0.05). The model of diagnosis for RCC constructed by the differential protein peaks (m/z 2945.35 、15340.8、6984.51 and 5819.23) generated excellent separation between the RCC and control groups. The sensitivity was 83.0% and the specificity of 84.6%. Blind test data indicated a sensitivity of 80.0 % and a specificity of 84.6%. Conclusions Differential protein peaks for RCC can be discovered in serum by technology combining magnetic beads with MALDI-TOF-MS. The pattern of combined markers provides a powerful and reliable diagnostic method for RCC with a high sensitivity and specificity.
