交联壳聚糖-碳纳米管复合物的制备及其对血红蛋白的吸附性能研究

摘要:采用表面沉积-交联方法制备了交联壳聚糖/多壁碳纳米管(Chit/MWCNTs)复合物,采用FTIR、SEM、TGA等技术手段对该复合物进行表征,结果表明,交联壳聚糖均匀地包覆于MWCNTs表面,其重量约为整个纳米复合物的35%。在pH=7.1的Tris-HCl缓冲介质中,该纳米复合物对血红蛋白表现出良好的吸附性能,经25min可达到吸附平衡,理论吸附容量为15.41 μg mg-1,比同等条件下交联壳聚糖的吸附性能提高近6倍。以此纳米复合物为吸附材料建立了血红蛋白的固相萃取分离方法,SDS-PAGE凝胶电泳结果证明以此纳米复合材料为固相吸附剂可有效地从人全血中分离出纯度较高的血红蛋白。

Investigations on the preparation of cross-linked Chit/MWCNTs nanocomposite and its adsorption of hemoglobin
Abstract:A chitosan/multi-walled carbon nano-tubes (Chit/MWCNTs) nanocomposite was prepared via surface deposition and cross linking methodology. It was characterized by FTIR, SEM and TGA analysis and the results indicated that the mass of the cross-linked chitosan layer coated on the surface of MWCNTs was about 35% of that of the total nanocomposite. In a medium of Tris-HCl buffer (pH7.1, 0.1mo L-1); the Chit/MWCNTs nanocomposite exhibited excellent adsorption capacity to hemoglobin and the adsorption equilibrium was reached within 25min. The maximum adsorption capacity was deduced to be 15.41 μg mg-1, i.e., a 6-flod improvement was achieved as compared to that obtained by pure cross-linked chitosan. A solid-phase extraction protocol for the separation of hemoglobin was then developed by using this nanocomposite as sorbent. The practical applicability of this system was demonstrated by processing human whole blood for the isolation of hemoglobin, and satisfactory results were obtained by assay with SDS-PAGE.
