超滤在猪血红蛋白纯化工艺中的应用研究

摘要:为了给人工红细胞的研究提供较高纯度和浓度的血红蛋白,本文采用超滤膜分离的方法,通过研究分析,在一定超滤条件,从新鲜猪血中分离纯化得到猪血红蛋白。并应用SDS-PAGE凝胶电泳、紫外-可见分光光度计对纯化后的血红蛋白进行了鉴定分析。经分析,优化的超滤条件为:操作压力0.05Mpa、料液温度12℃;在此条件下分离纯化的猪血红蛋白,纯度达到了99%以上,浓度较纯化前提高了约18.6倍。

Study on purification of porcine hemoglobin by ultrafiltration
Abstract:In order to obtain a high purity and concentration of porcine hemoglobin from the fresh swine blood to support the artificial red blood cell research, a ultrafiltration separating method was applied. The purified hemoglobin was analyzed by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) and ultra-visable spectrophotometer. The analysis and optimization of the ultrafiltration conditions is: operating pressure 0.05Mpa, feed temperature 12 ℃;And the results indicated that the purity of the porcine hemoglobin separated by ultrafiltration arrived a level of higher than 99% and the concentration of the hemoglobin was 18.6 times higher than before purification.
