丹参酮ⅡA与丹参酮ⅡA磺酸钠对K562细胞的增殖抑制与诱导分化作用

摘要:目的 比较丹参酮ⅡA与丹参酮ⅡA磺酸钠对人红白血病K562细胞的体外增殖抑制与诱导分化作用。 方法 采用改良MTT法测定两种药物对K562细胞的增殖抑制作用,倒置显微镜下观察其对K562细胞的形态学影响,以硝基四氮唑蓝还原试验法、流式细胞仪测定细胞膜表面分化抗原CD33和CD11b,观察丹参酮ⅡA与丹参酮ⅡA磺酸钠对K562细胞的诱导分化作用。 结果 3.4~13.6 μmoloL-1丹参酮ⅡA对K562细胞有显著的增殖抑制作用,且呈时间和剂量依赖性;丹参酮ⅡA对K562具有诱导分化作用,表现为NBT还原能力增强;CD33表达下降,CD11b表达升高。丹参酮ⅡA磺酸钠对K562细胞无增殖抑制与诱导分化作用。 结论 丹参酮ⅡA对K562细胞具有增殖抑制与诱导分化作用,其磺酰化后导致该药理活性丧失。

Inhibiting effect and inducing differentiation of tanshinone ⅡA and sodium tanshinone ⅡA sulfonate on K562 cells
Abstract:OBJECTIVE To investigate inhibiting effect and inducing differentiation of tanshinone ⅡA and sodium tanshinone ⅡA sulfonate on human erythroleukemia K562 cells in vitro. METHODS The inhibiting effect of tanshinone ⅡA and sodium tanshinone ⅡA sulfonate on the K562 cells were evaluated by modified MTT assay method, and the cell morphology were observed by inverted phase contrast microscope. The cell differentiation were evaluated by NBT reduction assay and measurements of CD33 and CD11b by flow cytometric analysis. RESULTS In tanshinone ⅡA group (3.4~13.6 μmoloL-1), the proliferation of K562 cells was significantly suppressed in a time- and dose- dependent manner, and K562 cells were differentiated into mature, which exhibited as increase on the NBT reducing capacity and the expression of CD11b, and decrease on the expression of CD33. However, neither growth inhibition nor differentiation induction was observed on K562 cells in sodium tanshinone ⅡA sulfonate group. CONCLUSION Tanshinone ⅡA showes a marked inhibition of cell proliferation and inducing differentiation on K562 cells but sodium tanshinone ⅡA sulfonate has no the anti-tumor effect to K562 cells.
