摘要:植物的核DNA含量和倍性水平是植物学研究中重要的基础研究指标。流式细胞仪作为检测这些指标的高效工具,在植物学研究的多个领域都发挥了重要作用。本实验室通过大量的植物流式细胞术实验,总结出一套详细通用的实验方法,同时本文阐述了各个实验环节的关键点,并分析了解决因碎片过多而导致实验失败的原因及解决办法,对今后进行植物流式实验具有指导意义。

Analysis of nuclear DNA content and ploid level in plant cells by flow cytometry
Abstract:Nuclear DNA content and ploidy level are key biological characters in the study of botany. As an efficient tool to measure them, flow cytometry has been playing very important role in many fields of botany research. Through plenty of flow cytometry experiments, our lab came up with a set of universal experimental procedure, at the same time, we have elaborated key points at every experimental stage and solutions to the problems caused by too much fragments which can result in the failure of an experiment, which may provide guidance for the future experiments.
