压力对共培养系统中气道平滑肌细胞增殖、形态及刚度的影响

摘要:目的 通过建立气道平滑肌细胞和气道上皮细胞间接共培养模型,为气道平滑肌细胞和气道上皮细胞之间的信息交流方式的研究提供借鉴与指导。方法 先分别单独培养大鼠气道平滑肌和上皮细胞,然后建立以聚对苯二甲酸乙二醇酯(PET)膜为介质的气道上皮细胞与气道平滑肌细胞联合培养模型。用倒置显微镜、MTT法和原子力显微镜(AFM)对四组不同模型中平滑肌细胞的生长情况和形态等进行研究。结果 在一定压力情况下,共培养模型中的气道上皮细胞对气道平滑肌细胞的增殖和刚度有促进作用。结论 受到压力刺激的情况下,研究共培养模型中气道上皮细胞对气道平滑肌细胞的影响,这对弄清细胞之间的信号传递方式及进一步明白哮喘的病理原因有重要的意义。

The impact of pressure to morphlogy,proliferation and rigidy of airway smooth muscle cell in co-culture system
Abstract:Objective Through the establishment of indirect co-culture model of airway smooth muscle cells and airway epithelial cells ,we can get reference and guidance to study the exchange methods of information between these two cells.. Methods Firstly, rat airway smooth muscle cells and epithelial cells were separately cultured, and then co-culture model of epithelial cells and smooth muscle cells was established by using polyethylene terephthalate (PET) membrane as medium. At last, the growth and morphology of smooth muscle cells on four different models were investigated by inverted microscope, MTT method and atomic force microscope (AFM). Results Under certain pressure, airway epithelial cells can promote the rigidity and proliferation of airway smooth muscle cells in the co-culture model. Conclusions The results indicate that airway epithelial cells cause the change of airway smooth muscle cells under the pressure, which is important to understand the signal way between cells and further study the pathology of asthma.
