基于紫外辐照改性聚苯乙烯膜固定化酶的目视比色法测定葡萄糖

摘要:通过简单的物理方法--紫外辐照,无需任何化学修饰,使疏水性的聚苯乙烯(PS) 膜表面形成亲水基团,从而获得分布均一、固定牢固的牛血清白蛋白包被层。以葡萄糖氧化酶作为酶模型,证实了经紫外辐照改性后PS膜可用于生物酶的有效固定,拓展了PS膜在酶生物传感器研究领域的应用。利用目视比色法、光电比色法研究了这种传感器的检测工作曲线、选择性、重现性及使用寿命,结果表明这种比色传感器可用于测定葡萄糖,且操作简便、成本低廉、制备简单,证实了聚苯乙烯可作为生物酶固定化的通用固载材料,为发展酶生物传感器提供一种普适性方法。

Visual colorimetric detection of Glucose based on UV-irradiated polystyrene membrane
Abstract:Using a simple physical method-ultraviolet irradiation, without any chemical modification, hydrophilic groups were generated and then Bovine serum albumin (BSA) was homogeneously modified on the polystyrene (PS) membrane. Glucose oxidase (GOD) used as enzyme model, a general immobilization method of enzyme based on UV-treated PS membrane had been demonstrated, which would popularize the application of PS carrier in the field of biosensor. Using visual colorimetric method, glucose with the concentration higher than 1 × 10-4 mol/L was successfully detected. Photoelectric colorimetric method had been also used to study sensitivity (LOD is 9.2 × 10-5 mol/L), selectivity, reproducibility and stability of the PS-based glucose biosensor.
