JNK信号转导通路对神经干细胞增殖的影响

摘要:目的 探讨JNK信号转导通路对新生大鼠海马神经干细胞增殖的影响。方法 采用无血清培养技术体外培养新生大鼠海马神经干细胞,传至第4代,大多数克隆球直径约为200 μm时,进行单细胞克隆培养,单细胞克隆形成的克隆球进行传代。将培养细胞分为2组,对照组采用神经干细胞培养液进行培养,实验组中在神经干细胞培养液的基础上添加不同浓度的JNK通路抑制剂SP600125, Western Blot检测加入细胞中JNK及p-JNK的表达情况, MTT法检测其对神经干细胞增殖的影响。结果 神经干细胞加入SP600125后JNK磷酸化水平明显低于对照组,当抑制剂浓度为10μM时细胞的增殖速度较对照组明显降低。2 w时全部死亡。结论 JNK信号转导通路被抑制后,神经干细胞的增殖能力明显降低。

Effects of JNK signaling pathway on the proliferation of neural stem cells
Abstract:Objective To study the effects of JNK pathway on the proliferation of neural stem cells from hippocampus of neonatal rats. Methods The hippocampus of neonatal rat was removed sterilely. NSCs were cultured in serum-free medium. Cell suspension was prepared and single cell clone was performed when the diameter of the fourth passage of clone sphere was 200μm. Monoclonal cells were passaged. NSCs were divided into control and experimental groups cultured in the mixture of NSCs media and inhibitor of JNK. The expression of JNK pathway was detected by western blot and the proliferation of NSCs was evaluated by MTT. Results The expression of phosphorylate JNK pathway in NSCs of the experimental group was lower than control group when the concentration of inhibitor was 10 uM, and NSCs proliferate slower than the control group. Conclusion The proliferation of neural stem cells significantly decreased when JNK signal transduction pathway was inhibited.
