达乌尔黄鼠(Spermophilus Dauricus)冬眠期骨骼肌氧化能力的研究

摘要:目的 研究不同时期达乌尔黄鼠骨骼肌呼吸酶活性和肌红蛋白含量的变化与其冬眠期抗肌萎缩机制间的关系。方法 黄鼠分冬眠前、冬眠一个月、冬眠两个月、出眠2天、出眠14天、春季组及夏季活跃组共7组,每组8只。用分光光度法测定比目鱼肌(Soleus,SOL)与趾长伸肌(Extensor digitorum longus,EDL)的柠檬酸合酶(有氧呼吸限速酶)、乳酸脱氢酶(无氧呼吸限速酶)及肌红蛋白的含量。结果 1)柠檬酸合酶活性与肌红蛋白的含量在SOL中表现出同步的周期性变化,冬眠期比冬眠前高,且出眠后继续增高,直到夏季结束,而在EDL中柠檬酸合酶在出眠2天时明显降低,出眠14天时略有回升,出眠一个月后恢复正常,各个时期的肌红蛋白含量无明显变化。2)冬眠黄鼠SOL的乳酸脱氢酶活性较冬眠前、出眠后均无统计学差异,EDL的乳酸脱氢酶活性,与冬眠前相比,亦无明显差异,但出眠2天和14天均有了明显上升。结论 骨骼肌中呼吸酶特别是有氧呼吸酶与肌红蛋白是黄鼠在冬眠期抗肌萎缩的重要作用因子,冬眠期黄鼠骨骼肌非凡的有氧呼吸能力可能是其抗肌萎缩的重要机制之一。

The Research on Oxidation Capacity of the Skeletal Muscle in Dauria Ground Squirrels(Spermophilus Dauricus) During Hibernation
Abstract:Objective: It was hypothesized that there was an mechanism of resisting muscle atrophy in dauria ground squirrels(Spermophilus Dauricus) .This assay was designed to study the mechanism by the measurement of the changes of myoglobin content and the activity of respiratory enzymes of the skeletal muscle. Methods: The ground squirrels were divided into seven groups: before hibernation, after 1 and 2 months of hibernation, after 2 and 14 days of arousal, spring group and summer group. Each group contained 8 animals. The content of citrate synthase, lactate dehydrogenase and myoglobin were measured by spectrophotometry. Results 1)The activity of citrate synthase and myoglobin content showed similar seasonal changes in soleus(SOL). In hibernation both of them were higher than that before hibernation and continued to increase after hibernation. The increase lasted to summer. Meanwhile the activity of the citrate synthase in extensor digitorum longus(EDL) decreased obviously after 2 days of arousal, then went up a little after 14 days of arousal, and came to normal in spring. Myoglobin content was almost the same during the different time in EDL. 2)There is no significant difference about the activity of lactate dehydrogenase in different season in SOL while it decreased significantly in the groups of before hibernation and hibernation in EDL. Conclusion:The extraordinary aerobic capacity is probably one of the important mechanism for its anti-atrophy. Myoglobin content and the activity of respiratory enzymes especially the aerobic ones are the key factor for the mechanism.
