St14-3-3蛋白在马铃薯储藏过程中的互作蛋白的捕获与分析

摘要:本研究克隆了一个在马铃薯块茎低温储藏过程中上调表达的14-3-3蛋白基因(St14-3-3),为了研究其在马铃薯低温糖化中的作用,将该蛋白在大肠杆菌BL21中诱导表达,通过亲和陷阱"捕获"了在低温糖化敏感的马铃薯栽培种E3与抗低温糖化的马铃薯野生材料(S. berthaultii)储藏过程中与之存在互作的蛋白质,并利用LC/MS 对蛋白质组分进行了分析,结果表明14-3-3蛋白的靶蛋白主要涉及到6-磷酸葡萄糖/磷酸转运(GPT2)、嘌呤的从头合成途径(磷酸核糖焦磷酸激酶4和磷酸核糖甲酰甘氨脒合酶)、蛋白质的合成正确折叠(热激蛋白HSP70等)以及细胞信号传导(硫氧还蛋白等)等,对靶蛋白所涉及到的代谢途径进行分析发现:磷酸戊糖途径可能参与到了马铃薯低温糖化的调节。

Capture and analysis of interacted proteins of St14-3-3 during the colg storage of potato tubers
Abstract:A 14-3-3 protein (St14-3-3) up-regulated in early-stage of cold storage of potato tubers was cloned in this research. To figure out the function of St14-3-3 in low temperature sweetening, interacted proteins during the storage of a LTS-resistant wild potato species(S. berthaultii) and LTS-sensitive potato cv. E3 tubers were "captured" by affinity trapping, and the components of obtained proteins from LTS-resistant poatoto (S. berthaultii) tubers stored at 4℃ for 10d were analyzed by LC/MS. The data showed that target proteins of St14-3-3 were involved in Glucose-6-phosphate transport (GPT2), de novo synthesis of purine (KPRS4 and PUR4), protein synthesis and folding (HSP70, et al), cellular signaling (thioredoxin, et al) and so on. Analysis of the pathways involved suggests that oxidative pentose phosphate pathway may be involved in regulation of LTS.
