摘要:炎症复合物(inflammasome)是细胞内由多个蛋白组成的分子反应平台,细胞感染或外界压力能将它激活,它通过活化半胱天冬酶-1来调控白介素-1β,白介素-18等促炎细胞因子的成熟,启动天然免疫防御反应。炎症复合物活性的失调与许多人类炎症疾病有关。这里将以NLRP1,NLRP3,IPAF,AIM2炎症复合物的组成、功能、分子调控机制为重点,详细介绍炎症复合物及相关细胞信号转导研究的最新进展及其在治疗多种人类免疫疾病中的重要意义。

Research progress on inflammasome and related cell signaling
Abstract:Inflammasomes are multiprotein complexes and caspase-1-activating platforms. Upon cellular infection or stress, inflammasomes are activated and they trigger the maturation of proinflammatory cytokines such as interleukin-1β, -18 to initiate innate immune defenses. The dysfunction of inflammasomes is proved to be associated with many human inflammatory diseases. The current research progress on inflammasomes and related cell signaling, as well as its important role in treatment of human inflammatory diseases, is reviewed here. In particular, the components, functions and regulatory mechanisms of the NLRP1, NLRP3, IPAF and AIM2 inflammasomes are discussed.
