冬眠及后肢去负荷对达乌尔黄鼠骨骼肌功能的影响

摘要:目的 研究达乌尔黄鼠(Spermophilus dauricus,简称黄鼠)在冬眠期间及后肢去负荷状态骨骼肌收缩功能的变化,探讨与废用性肌萎缩的关系。方法 (1) 将黄鼠分为冬眠前、冬眠中、出眠和后肢去负荷组进行各指标的观测;后肢去负荷模型参考大鼠尾部悬吊法建立,尾部悬吊14天后进行各指标检测,其对照组为冬眠前组;(2). 采用电子刺激系统和Powerlab系统采集、分析骨骼肌收缩功能相关数据;定磷法测定骨骼肌钙ATP酶活性。结果 (1).在冬眠中,比目鱼肌、趾长伸肌的单收缩最大张力(Pt)、强直收缩最大张力(P0)均显著降低;达到最大收缩张力50%的时间(TP50)、达到最大收缩张力的时间(TPT)、舒张至最大收缩张力50%的时间(RT50)以及舒张至最大收缩张力75%的时间(RT75)均显著增加;比目鱼肌、趾长伸肌的钙ATP酶活性没有显著变化。(2).出眠时,各指标与冬眠前相比均无显著差异。(3). 后肢去负荷14天后,比目鱼肌和趾长伸肌的Pt、P0、TP50、TPT、RT50、RT75及钙ATP酶活性均无显著变化。结论 (1)冬眠中黄鼠比目鱼肌和趾长伸肌钙ATP酶活性保持稳定,表明其肌肉功能未受明显影响,而骨骼肌收缩张力下降、收缩相和舒张相延长可能是由于骨骼肌纤维类型由快缩型向慢缩型转化所引起。(2)尾部悬吊14天不会引起黄鼠明显的废用性肌萎缩。

Hibernation and Hindlimb-unloading Effect on the Skeletal Muscle in Ground Squirrels
Abstract:Objectives: To study the changes of skeletal muscle function of the ground squirrel (Spermophilus dauricus) during hibernation and under the condition of the hindlimb-unloading. Methods: (1).Divide ground squirrels into 4 groups: pre-hibernation (Pre-H), hibernation (H), post-hibernation (Post-H) and hindlimb-unloading (HU); the hindlimb-unloading model was constructed by tail suspension of the ground squirrel for 14-day according to the tail-suspension model of rat, and it's control group is the Pre-H group. (2). Collect and analyze the data of skeletal muscle funcion through the Powerlab system; the UV spectrophotometer was used to detect the Ca2+-ATPase's activity according to the Pi content. Results: (1). During the hibernation, the peak twitch tension (Pt) and peak tetanic tension (P0) of both soleus(SOL) and extensor digitorum longus(EDL) have decreased significantly; the time to 50% peak tension (TP50), time to peak tension (TPT), time from peak tension to 50% relaxation (RT50) and time from peak tension to 75% relaxation (RT75) have incerased notably; the Ca2+-ATPase's activity of the SOL and EDL has no significant changes during the hibernation. (2). During the period of post-hibernation (Post-H), all the indicators seemed recovered as same as the pre-hibernation. (3). After 14-day-hindlimb unloading, no changes was found about the Pt, P0, TP50, TPT, RT50, RT75 and Ca2+-ATPase's activity of both SOL and EDL. Conclusions: (1). Since the Ca2+-ATPase's activity of both SOL and EDL kept stability, the decreas of the muscle tension and the increase of the time course of SOL and EDL of the ground squirrel may be due to the transition of the muscle fiber type: from the fast twitch fiber to the slow twitch fiber. (2). The 14-day-hindlimb-unloading can not induce a significant disuse atrophy in the ground squirrel.
