水稻黄胚乳突变体JMW的基因定位与克隆

摘要:乙醛脱氢酶(ALDH)是生物界中常见的一种酶类,依赖于辅酶NAD(P)+,乙醛脱氢酶负责把代谢中各种醛类氧化成对应的羧酸,植物中已发现的乙醛脱氢酶的表达大多数与渗透压逆境相关。本研究从一个水稻低谷蛋白品种(W3660)中,鉴定出一个天然的黄色胚乳突变体,命名为jmw(金米王),突变体色素的积累主要发生在种子发育的后期,并且受发育时期和储存时间的影响。用图位克隆和转基因的方法发现是由于OsALDH7基因单碱基突变,造成蛋白翻译的提前终止,从而产生突变体表型,OsALDH7的表达模式与前人研究相似。另外我们也发现,不同家族的乙醛脱氢酶之间有一定的功能冗余。本研究对进一步阐述OsALDH7基因的功能奠定了良好的基础。

Gene Mapping and Cloning of a Yellow Endosperm Mutant in Rice (Oryza sativa L.)
Abstract:Aldehyde dehydrogenase (ALDH) represents a superfamily of NAD(P)+ -dependent enzymes that oxidize aldehydes into their corresponding carboxylic acids. Most of ALDHs in plants were found to be associated with osmotic stress. In this study, a natural mutant of yellow endosperm was identified from a low-glutelin var. W3660, which named as jmw (new golden rice in Chinese). The pigment was mainly accumulated in the later stage of seed maturation, and was associated with developmental and storage period. Map-based cloning and complement experiment indicated that the mutant of OsALDH7 was responsible for the mutant phenotype, and the expression pattern of OsALDH7 was similar to the pervious studies. We also found the possible compensating pattern between ALDH families. The present study laid a firm foundation on the further functional study of OsALDH7.
