棉花ADF基因克隆鉴定与差异表达分析

摘要:肌动蛋白解聚合因子(actin-depolymerizing factor, ADF)是一种在真核生物中广泛存在的低分子量的肌动蛋白结合蛋白,它在调控细胞内肌动蛋白纤丝的解聚合和再聚合中起着关键作用。我们在棉纤维cDNA文库中分离克隆了7个ADF基因(cDNAs),分别命名为GhADF2,GhADF3,GhADF4,GhADF5, GhADF6,GhADF7,GhADF8。RT-PCR分析表明,GhADF2在纤维中优势表达,而GhADF5基因则在子叶中表达量最高。GhADF6 和 GhADF8 在花瓣中优势表达,而GhADF7 在花药中特异表达。GhADF7 转录本在开花当天的花药中达到最大值,表明该基因受花药发育调节。另一方面,GhADF3和GhADF4似乎不具有组织特异性或偏爱性表达。同一组织中不同GhADF基因表达量有较大的差异,表明它们可能涉及棉花不同组织生长发育过程的调节。而且,在进化过程中,各ADF同分异构体之间可能发展形成某种功能上的差异性。

Molecular characterization of seven ADF genes differentially expressed in cotton
Abstract:ADF (actin depolymerizing factor), highly conserved in all eukaryotic cells, is a low molecular mass of actin-binding protein. It plays a key role in modulating the polymerizing and depolymerizing of the actin filaments. Seven cDNAs (designated GhADF2, GhADF3, GhADF4, GhADF5, GhADF6, GhADF7 and GhADF8, respectively) encoding ADF proteins were isolated from cotton cDNA libraries. GhADF2/3/6/7/8 encode protein of 139 amino acids, GhADF4 deduces a protein with 143 amino acids, and GhADF5 encodes a protein of 141 amino acids. Real-time quantitative RT-PCR analysis showed that GhADF2 is predominantly expressed in fiber, whereas GhADF5 is mainly expressed in cotyledon. GhADF6 and GhADF8 were preferentially expressed in petals, whereas GhADF7 displayed the highest level of expression in anther, revealing its anther specificity. The GhADF7 transcripts in anther reached its peak value at flowering, suggesting that its expression is developmentally-regulated in anther. On the other hand, GhADF3 and GhADF4 are likely to have no tissue specificity. The expression levels of different ADF genes may vary considerably in same cell type, suggesting that they may be involved in regulating tissue development of cotton and the each ADF isoform may develop the functional difference during evolution.
