辐射诱导ATF3表达特性及调控研究

摘要:目的:研究电离辐射诱导应激反应基因ATF3表达规律及其调控机制,为辐射损伤分子机理研究寻找新的途径。方法:应用Westesn Blot观察辐射诱导ATF3蛋白表达规律。利用Cat-assay技术研究ATF3在启动水平上的调控。RT-PCR分析ATF3转录调控。结果:电离辐射在一定剂量范围内可诱导ATF3蛋白表达。不用剂量辐射照射对于ATF3基因启动子的激活没有影响。ATF3蛋白表达水平的提高的同时,RNA表达水平明显增高。结论:辐射诱导的ATF3蛋白表达具有明显的时效性变化和剂量效应变化规律。ATF3蛋白表达调控发生在RNA转录水平,而与ATF3基因启动子激活无关。

Radiation-induced expression and regulation of ATF3
Abstract:Objective: This work studies the stress response induced by ionizing radiation and regulation of gene expression pattern of ATF3 mechanism and the molecular mechanism of radiation damage. Methods: Westesn Blot induced ATF3 protein expression is observed for revealing mechanism of radiation. Cat-assay technique is used for ATF3 regulation of the level at startup. RT-PCR analysis has been used for ATF3 transcription. Results: The dose of ionizing radiation in a certain range of protein expression can be induced by ATF3. No dose of radiation for the activation of ATF3 gene promoter has no effect. ATF3 protein levels increase and RNA expression was increased. Conclusion: The radiation-induced ATF3 protein expression changes with significant time and dose effects were studied. Regulation of ATF3 protein expression levels in RNA transcription, but not related to activation of ATF3.
