创伤后应激障碍大鼠中缝背核5-HT1A受体的改变

摘要:目的 观察创伤后应激障碍(PTSD)样行为异常大鼠中缝背核(the dorsal raphe nucleus,DRN)神经元5-HT1A受体表达变化,有望揭示PTSD的部分发病机制。方法 成年健康雄性Wistar大鼠60只,随机分为连续单一刺激(single prolonged stress, SPS)模型的1d、4 d、7d组及正常对照组,采用免疫组化、Western blott等方法,检测PTSD样行为异常大鼠中缝背核神经元5-HT1A受体表达变化。结果 SPS刺激后大鼠中缝背核神经元5-HT1A受体于1d 4d,7d逐渐升高,高于对照组。结论 大鼠中缝背核神经元5-HT1A受体表达变化,可能与PTSD样大鼠恐惧增强的发病机制相关。

Changes of 5-HT1A receptor in the dorsal raphe nucleus in the rat model of post-traumatic stress disorder
Abstract:IObjective To observe the changes of the 5-HT1A receptor expression in the dorsal raphe nucleus of PTSD rats to reveal part of the pathogensi. Methods A total of 60 male Wistar rats were randomly divided into 1d, 4d and 7d groups of SPS and a normal control group. Immunohistochemical and Western blot techniques were used to observe and detect the changes in the expression of 5-HT1A receptor(5-HT1AR) in the dorsal raphe nucleus, and image analysis and statistical analysis were performed. Results The expression of 5-HT1AR was sharply increased at 1d, and gradually incresed in 4d,7d by compared with control group. Conclusion These results suggest that the changes in the expression of 5-HT1AR in the dorsal raphe nucleus may be related to the pathogenesis of enhanced fear in PTSD rats.
