斑马鱼血管中致动脉粥样硬化脂质的浓度极化现象

摘要:低密度脂蛋白(low density lipoprotein, LDL)浓度极化是动脉粥样硬化发生发展过程中的重要刺激因素。但是,到目前为止还未见脂质浓度极化在体研究的相关报道。本研究以转基因斑马鱼(Flk1:GFP)为模型,在其受精后第二天(2dpf),将红色荧光标记的低密度脂蛋白(Dil-LDL)显微注射到血液循环系统内(斑马鱼注射总量n=60)。经激光共聚焦显微镜跟踪脂质在血管中的流动、分布以及沉积情况,再应用图像处理软件对获取的图片进行荧光定量分析。结果显示:与对照组相比,注射组(57/60)血管内有红色脂蛋白在血液中流动,统计分析显示血液与血管壁界面的低密度脂蛋白浓度显著高于血流中的本体浓度。注射两天后,出现明显的脂质沉积,并且主要出现在血管的分叉和弯曲处。研究结果显示斑马鱼模型适合做脂质沉积的在体研究,并证实在脉管系统中确实存在脂质浓度极化现象。

Concentration Polarization of Atherogenic Lipids in Vessel in Zebrafish
Abstract:The concentration polarization of low- density lipoproteins plays a crucial role in the genesis and progression of atherosclerosis. However, it is few to investigate the relations between them in vivo. LDL with green fluorescence was injected into optically transparent Flk1:mCherry Ras zebrafish larvae(Day 2,n=60). The flow, distribution and deposition of the lipids in the vessel were observed by confocal microscope, and then the images were analyzed by fluorescence analysis software. The results show that the green fluorescent lipoproteins are flowing in the vessel luminal (57/60), compared to the control with no green fluorescent proteins, using confocal microscopy. LDL is accumulated 2 days after injection, and there are amount of deposition in the vascular bifurcation and bend. According the in vivo research confirmed that the lipoprotein concentration polarization was occurred in vasculature.
