大豆苷原对去势大鼠子宫及乳腺的选择性作用及其ER和PPARγ表达的影响

摘要:目的:探讨大豆苷原对子宫内膜和乳腺组织的选择性作用及其机制。方法:将4月龄大鼠随机分为6组:假手术组(Sham)、去势组(OVX)、去势+戊酸雌二醇组(E2,戊酸雌二醇800 μgokg-1od-1)、去势+高剂量大豆苷原组(H-Dai,大豆苷原200 mgokg-1od-1)、去势+中剂量大豆苷原组(M-Dai,大豆苷原50 mgokg-1od-1)和去势+低剂量大豆苷原组(L-Dai,大豆苷原10 mgokg-1od-1)。除假手术组外,其余大鼠均切除卵巢,术后4周开始以相应的药物灌胃,3个月后处死大鼠。取大鼠子宫及乳腺组织做相应检测。结果:H-Dai组大鼠子宫内膜腔上皮、间质和内膜厚度显著厚于OVX组(P<0.01)和L-Dai组(P<0.05)。乳腺腺泡计数Sham组大鼠为9.1个,E2组为2.3个,其余组未见明显腺泡。H-Dai组大鼠子宫内膜ERα mRNA表达显著高于OVX组(P<0.01),且高于L-Dai组。乳腺组织ERα表达差异无统计学意义。ERβ在3个剂量Dai组大鼠子宫内膜和乳腺组织均未检测出,3个剂量Dai组之间PPARγ差异无统计学意义。结论:大豆苷原对去势大鼠子宫内膜和乳腺组织具有选择性作用。高剂量大豆苷原可能通过刺激子宫内膜ERα,对内膜产生弱雌激素样作用,中、低剂量则无刺激性作用。大豆苷原对乳腺组织则无刺激作用。

The Tissue-selective Effect of Daidzein On Uterus and Lacteal Gland and the Expression of Estrogen Receptors and Peroxisome Proliferator-activated Receptor Gamma of the Ovariectomized Rats
Abstract:Objective: To investigate the tissue-selective effect of daidzein on uterus and lacteal gland and it's molecular mechanisms. Materials and methods: Sixty four-month-old Sprague-Dawley(SD)female rats were allotted to one of six groups. Four weeks after reproducing the osteoporosis model of ovariectomized SD rats, Sham group and control OVX were administrated with distilled water, whereas the other groups received valerate estriol(800 μgokg-1od-1)and different doses of daidzein(10,50,200 mgokg-1od-1)by gavage respectively for three months. After the sacrifice of the rats, we detected histomorphometry of uterine endometrial and lacteal gland tissue, and the expressions of mRNA of ERα, ERβ and PPARγ realtime-RT-PCR, while protein were by immunohistochemistry in them. Results: Endometrial luminal epithelium, stroma, and endometrial thickness of rats in H-Dai group were significantly thicker than that of OVX group (P<0.01) and L-Dai group (P<0.05). Lacteal gland of rats in Sham group was counted as 9.1, while E2 group was 2.3, no clear gland in other groups. The expression of ERα mRNA of rats' endometrial in H-Dai group was significantly higher than that of OVX group (P<0.01), and higher than that of L-Dai group. The expression of ERα in breast tissue was no significant difference. The expression of ERβ rats' endometrial and breast tissue in three Dai groups were not detected, and the expression of PPARγ was not significant different among three Dai groups. Conclusion:There was tissue-selective effect of daidzein on uterus and lacteal gland of the ovariectomized rats. High-dose daidzein may play weak estrogen-like effect on uterine endometrial tissue through ERα, while no estrogen-like stimulation on breast.
