PNGF复合材料对细胞外调节蛋白激酶磷酸化的影响

摘要:为进一步研究复合材料PNGF修复周围神经组织的机理,将该材料的浸提液与大鼠神经嵴源性PC12细胞共培养后,经细胞总蛋白提取,且通过ELISA试剂盒和westernblot实验分析与PC12细胞分化相关的细胞外调节蛋白激酶(简称ERK1/2)磷酸化的表达量,探索浸提液微环境对细胞诱导过程的影响。其结果表明:在材料中复合神经生长因子NGF后,该复合材料的浸提液在早期能促进PC12细胞ERK1/2蛋白磷酸化水平明显提高,并最终诱导细胞分化。?????

Influence of PNGF composite materials to the extracellular regulated protein phosphorylation
Abstract:For further research on the mechanism of PNGF composite materials to repair peripheral nerve tissue. Culture PC12 cells with the PNGF material extracting liquid, Enzyme-linked immunosorbent assay(ELISA) and westernblot experiment were used to detect the quantity of ERK1/2 phosphorylation expression. The results showed that the nerve growth factor of the composite material which released from the degradable material after being extracted can promote the extracellular regulated protein(ERK1 1/2) phosphorylation level obviously in PC12 cells early, and at last the leaching carry liquid of PNGF composite materials will induce PC12 cells differentiation.?????
