产β-葡萄糖苷酶的菌种的筛选,鉴定及其酶学特性的研究

摘要:本文从土样中分离筛选出一种活性较高的产β-葡萄糖苷酶的菌种WGEA1,并对其进行初步的鉴定和采用DNS法测其粗酶液的酶学特性。酶学特性表明,WGEA1最适生长温度是在50℃-55℃之间,最适PH在6-7之间;而在低于50℃条件下,WGEA1产酶的PH为5-8时,酶活性较稳定,在最适反应时间30min下,金属离子和有机溶剂都对酶活性影响很大,这些发现都将为下一步的研究酶法合成烷基糖苷奠定了基础。

The screening and identification of the strains producing β-glucosidase,and properties of β-glucosidase
Abstract:One strain of WGEA1 producing higher activity of β-glucosidase,were isolated from soil samples, and then preliminarily identified of the strain and measured the enzymatic properties of crude enzyme . The enzymatic properties showed that the optimum temperature of WGEA1 is between 50℃ and 55℃,the optimum PH is between 6 and 7;In less then 50℃, the activity of β-glucosidase from WGEA1 is stable for the PH 5-8, At the optimum reaction time of 30 min,the metal ions and organic solvents have a great influence on the activity of β-glucosidase,and the next step for the research of the enzymatic synthesis of alkyl glycoside laid the foundation.
