纳米金溶胶细胞毒性的研究

摘要:本文主要研究纳米金溶胶的细胞毒性。通过[3H]-TdR掺入法研究了粒径在15-20nm的纳米金溶胶,溶胶中含有的柠檬酸钠溶液,以及溶胶中含有的柠檬酸钠与PVP混合溶液对正常细胞(人体表皮细胞、人体皮肤成纤维细胞)和癌细胞(HeLa细胞、K562细胞)活性的影响。实验结果为当浓度小于50μmol/L时,随浓度的增加,其对正常细胞增殖的促进作用增加;而后随浓度的增加促进作用减弱,并逐步转化为对正常细胞增殖的抑制作用。当纳米金的浓度不大于50μmol/L时,纳米金对癌细胞的细胞活性没有明显影响;浓度大于50μmol/L时,随浓度的增加,对癌细胞抑制作用增强。由此可以看出,纳米金溶胶中含有的柠檬酸钠与PVP对这四种细胞的活性基本没有影响,可以在一定浓度范围内促进人体正常皮肤细胞的增殖,具有剂量依赖性和时间依赖性;对于癌细胞的影响,则需高于一定浓度(50μmol/L)才有显著的抑制作用。

Study on the Cytotoxicity of Gold Nanoparticle Colloids
Abstract:In this paper,we aim to study the cytotoxicity of gold nanoparticle colloids. We studied the effect of gold nanoparticle colloids of diameter 15nm to 20nm with sodium citrate solution and those with the mixture of sodium citrate and Poly Vingl Pyrrolidone (PVP) solution to the activity of normal cells of Human skin(human epidermal cell, Human hypertrophic scar derived fibroblasts also called HSF) and cancer cells(HeLa, K562, CHO) by 3H-TdR incorporation method. Results demonstrate that gold nano-particle colloids within a certain concentration range could promote the proliferation of normal cells of Human Skin in a time-dependent and dose-dependent manner. If the concentration was below 50μmol / L, the facilitation of gold nanoparticle colloids towards cell proliferation strengthened as the concentration increased. At 50μmol / L, the facilitation ability was most significant. However, the facilitation ability decreased as the concentration continually increased above 50μmol / L, and gradually converted to inhibitive effect. The activity of cancer cells showed invisible changes when the concentration of Gold nano-particle colloids was less than 50μmol / L, and when the concentration was above 50μmol / L, the inhibitive effect on cancer cell increased as the concentration increased.
