多亮氨酸重复区免疫球蛋白样蛋白在成年大鼠脑组织的表达及其意义

摘要:目的:观察多亮氨酸重复区免疫球蛋白样蛋白(leucine-rich and immunoglobulin-like domains, LRIG)在成年大鼠脑组织中的表达,根据其表达分布及形态对LRIG1,LRIG2,LRIG3在神经系统的功能进行初步探讨。方法:无RNA酶条件下制作正常成年大鼠脑组织石蜡切片;设计、制作LRIG1,LRIG2,LRIG3 mRNA寡核苷酸探针,原位杂交检测LRIG1,LRIG2,LRIG3在成年大鼠脑组织中的表达,从组织形态学角度进行观察和研究。结果:LRIG1,LRIG2,LRIG3在成年大鼠大脑皮质中均有表达,但在皮层各层分布不同;神经元胞体均可见LRIG1,LRIG2,LRIG3表达。海马CA1区及齿状回颗粒细胞下层可检测到LRIG1高表达。结论:LRIG1,LRIG2,LRIG3可能参与了中枢神经系统的高级功能,LRIG1可能参与了海马学习和记忆的功能,及成年大鼠海马齿状回神经发生的过程。

Expression pattern of LRIGs in adult rat brain
Abstract:To investigate the expression pattern of leucine-rich and immunoglobulin-like domains (LRIG)proteins in the brain of adult rat and preliminarily explore the function of LRIGs in the central nervous system. Methods: The oligonucleotide probes of LRIG1, LRIG2, and LRIG3 were designed and synthesized. The tissue sections of adult rat brain were manufactured under RNase-free condition, followed by in situ hybridization with the LRIGs probes. The possible functions of LRIGs in the central nervous system were preliminarily discussed according to the expression pattern and the distribution of LRIGs. Results: The three proteins of LRIG1, LRIG2, and LRIG3 were detected to be expressing in the cerebral cortex. LRIG1 was also observed to be expressing in the CA1 region and the subgranular zone of hippocampus. Conclusion: LRIG1, LRIG2, and LRIG3 may all play some roles in advanced brain function of adult rats. LRIG1 may be involved into the neurogenesis of adult rats.
