右美沙芬制剂的药代动力学研究

摘要:目的 建立高效液相色谱法测定血浆中右美沙芬的浓度,研究右美沙芬咀嚼胶片的药动学和生物利用度。方法 以市售右美沙芬咀嚼片为对照品,运用3P37药动学软件处理数据,进行药动学和生物利用度研究。 结果 右美沙芬咀嚼胶片AUC(0→∞、Cmax和tmax分别为(488.76 ± 175.00) ngomL-1.h、(95.45 ± 17.53) ngomL-1、(1.83 ± 0.57) h;市售咀嚼片AUC(0→∞)、Cmax)和tmax分别为(370.13 ± 90.56) ngomL-1.h、(174.00 ± 47.88) ngomL-1、(1.04 ± 0.14) h。咀嚼胶片的相对生物利用度为(140.73 ± 65.91)%。 结论 相对于常规咀嚼片,咀嚼胶片AUC(0→∞)有所提高,相对生物利用度较高,Tmax显著延长,但Cmax降低。

Studies on pharmacokinetics of new dextromethorphan preparation
Abstract:IObjective To establish an HPLC method for the analysis of pharmacokinetics and relative bioavailability of the chewing gum medicine tablet after oral administration in rabbit. Methods The pharmacokinetics parameters and the relative bioavailability of the new chewing gum medicine system compared with chewing tablet were computed by software 3P97. Results The main pharmacokinetics parameters of the new chewing gum medicine system were as follows: AUC (488.76 ± 175.00) ngomL-1.h, Cmax(95.45 ± 17.53) ngomL-1, tmax(1.83 ± 0.57) h. The main pharmacokinetics parameters of the chewing tablet were as follows: AUC (370.13 ± 90.56) ngomL-1.h, Cmax(174.00 ± 47.88) ngomL-1, tmax(1.04 ± 0.14) h. The relative bioavailability of the new chewing gum medicine system was (140.73 ± 65.91)%. Conclusion For the new chewing gum medicine system, its AUC(0→∞)was increased, but there are no significant differences; Cmax decreased;tmax prolonged;the relative bioavailability of the new chewing gum medicine system was greatly enhanced.
