紫外-荧光微孔板酶检测技术测定土壤酶活性

摘要:采用一种新的土壤酶检测技术-荧光微孔板酶检测技术,测定土壤硫酸酯酶、磷酸酶、?-葡萄糖苷酶和肽酶活性,采用紫外微孔板酶检测技术测定多酚氧化酶和过氧化物酶活性。结果表明,紫外-荧光微孔板酶检测技术可快速测定土壤中酶的活性。采自农田的昔格达土的硫酸酯酶、磷酸酶和肽酶活性高于紫色土和采自矿山的昔格达土的相应的酶活性;矿山昔格达土的多酚氧化酶和过氧化物酶活性在三者中最高;紫色土的β-葡萄糖苷酶活性高于昔格达土的β-葡萄糖苷酶活性。

Enzyme Assay of Soil using Ultraviolet and Fluorescence Microplate
Abstract:The activity of soil sulphatase, phosphatase, peptidase and -glucosidase were measured using 96-well microplate fluorimetric assay, and phenol oxidase and peroxidase activity were measured using 96-well microplate ultraviolet spectrophotometry. The correlation between soil physical and chemical properties and enzyme activities were discussed. The activity of sulphatase, phosphatase and peptidase of Xigeda soil taken from farmland were higher than Xigeda soil taken from mining area and purple soil; -glucosidase activity of purple soil was the highest in tested soils; but phenol oxidase and peroxidase activity of Xigeda soil taken from mining area were the highest.
