顺铂-羟基磷灰石-壳聚糖缓释微球的合成及其性状研究

摘要:目的 制备顺铂-纳米羟基磷灰石-壳聚糖(DDP-HAP-CS)缓释微球,观察其性状和载药率。方法 以纳米羟基磷灰石(HAP)、壳聚糖(CS)和顺铂(DDP)为原料,采用乳化交联法制备DDP-HAP-CS缓释微球;采用扫描电子显微镜观察微球的大小和表面形貌,X射线衍射仪分析交联后微球的物相及其结晶程度;原子吸收分光光度计测定缓释微球中DDP的含量,动态渗析法检测该缓释微球DDP体外释放曲线。结果 成功制备了DDP-HAP-CS缓释微球,直径约为5μm-20μm,微球中主要为弱结晶CS和HAP晶相。微球中DDP的含量为16.46%,该缓释微球DDP体外释放释放率随时间延长而增加,24h内DDP的释放率增加较快,24h-72h释放率处于平台期,72h后释放率又开始缓慢上升,至120h后重新进入平台期。结论 DDP-HAP-CS缓释微球可以使DDP在体外条件下缓慢释放,基本达到了缓释药物的要求。

Experimental Study of Synthesis and Characteristics of Cisplantin- nano Hydroxyapatite-Chitosan Microspheres
Abstract:IObjective To prepare Cisplantin-nano Hydroxyapatite-Chitosan Microspheres, and investigate its characteristics. Methods Nano-hydroxyapatite (HAP), chitosan (CS) and cisplatin (DDP) were as raw materials, and DDP-HAP-CS release microspheres were prepared by emulsion cross-linked method. The size and surface morphology of microspheres were observed by scanning electron microscopy, and the phase and crystallinity of microspheres were detected by X-ray diffraction analysis. DDP content of microspheres was detected by atomic absorption spectrophotometer, and the DDP release profile of microspheres was detected by dynamic dialysis method. Results DDP-HAP-CS release microspheres were prepared with diameter of 5μm-20μm, and DDP content of microspheres was 16.46%. The release rate of DDP from microspheres increased with time in vitro. The release rate of DDP increased rapidly in the first 24h, and the release rate reached plateau stage in 24-72h, then the release rate rose slowly, and reached plateau stage again after 120h. Conclusion DDP-HAP-CS release microspheres can make DDP slow release in vitro, it can meet the basic requirement of sustained release of drugs.
