阻塞性黄疸大鼠核受体FXR运动干预下的表达研究

摘要:目的:通过中小强度有氧运动干预,观察运动7天和14天后大鼠阻塞性黄疸致轻度肝损伤的FXR mRNA表达。方法:36只SD大鼠随机分为阻塞性黄疸、早期运动及晚期运动组,运动组进行坡度5%、速度12.0 m/min、每天30 min(18~19时之间),共训练14天的中小强度跑台训练。运动结束后大鼠禁食12小时,随后分离出肝组织并取材,应用HE染色法观察大鼠肝细胞形态;生物化学法测定血清总胆红素(TBIL)、谷丙转氨酶(ALT)、谷草转氨酶(AST)含量;RT-PCR技术检测FXR、β-actin mRNA表达。结果:HE染色结果显示,阻塞性黄疸组可见肝细胞广泛空泡样变性、片状坏死,早期运动组肝细胞少量点状坏死,晚期运动组肝细胞未见明显坏死;血清学检查结果显示,早期运动组和晚期运动组大鼠血清TBIL、AST、ALT含量显著低于阻塞性黄疸致肝损伤组(P < 0.05);RT-PCR检测结果显示,早期运动组合晚期运动组大鼠FXR mRNA表达显著高于阻塞性黄疸致肝损伤组(P<0.05)。结论:中小强度有氧运动对阻黄导致的轻度肝损伤有较好的效果,其机制可能通过促进FXR表达与增强胆汁酸的肠肝循环及排泄,从而发挥肝脏的自我修复作用。

Obstructive jaundice nuclear receptor FXR exercise intervention with expression study in the rats
Abstract:Objective: Through the small or medium strength aerobic exercise intervention, observed 7 days and 14 days of obstructive jaundice rats to mild liver injury FXR mRNA expressions. Methods: 36 SD rats were divided into obstructive jaundice, early exercise and later exercise interventions 3 groups, exercise group speed by 5%, and the slope of 12.0 m/min, thirty minutes a day, trained 14 days of small or medium strength training run sets. Exercise end rats after fasting 12 hours, and then separating out the liver tissue and materials, HE staining of liver cell morphology change; biochemical analysis methods of serum total bilirubin (TBIL),glutamic-pyruvic transaminase (ALT) and glutamic-oxaloacetic transaminase (AST) contents; RT-PCR detection of FXR and β-actin mRNA expression levels. Result: HE staining of liver cells in the obstructive jaundice group showed extensive vacuolar degeneration of liver cells, necrosis, early exercise group showed a small amount of spotty necrosis of liver cells, later exercise group not have obvious cell necrosis; serum levels examination showed early exercise group and later exercise group SD serum TBIL, AST and ALT content than the obstructive jaundice liver damage group content is low (P<0.05); RT-PCR assay techniques showed early exercise group and later exercise group compared with SD rats EGF mRNA expression level was higher obstructive jaundice(P<0.05). Conclusion : Small or medium strength aerobic exercise for resistance to damage the liver of obstructive jaundice have a good effects, its mechanism may enhanced FXR expression and promoted the bowel bile acid enterohepatic circulation and excretion, play a role of liver repair itself.
