mRNA 结合蛋白Vigilin功能初探

摘要:Vigilin 是一个RNA结合蛋白,由 14 个连续的RNA结合结构域--- KH 结构域组成。Vigilin 可以通过与 mRNA 3’UTR 结合调控 mRNA 的翻译。本文通过 EMSA 实验证明了 Vigilin 结合于 ER-beta 3’UTR 。双荧光素酶报告基因实验发现高表达 Vigilin 可以提高 ER-beta 的表达,利用用带荧光蛋白标签的融合蛋白进行的共定位实验发现 Vigilin 与去帽酶 Dcp1B 有共定位现象,而 Dcp1B 是 mRNA 降解过程中重要的一环,说明 Vigilin 与 mRNA 降解相关。

Function of mRNA Binding Protein Vigilin
Abstract:Vigilin is known as an RNA binding protein, which consist of 14 serial RNA binding domain---KH domain. Vigilin was reported to combine to mRNA 3’ UTR and regulate posttranscription which mechanism is unknown yet. This study use EMSA confirmed Vigilin binding to ER-beta 3’ UTR which was first found by our lab earlier , and applied Dual-luciferase Assay found out overexpression of Vigilin can raise the expression of ER-beta; also fluorescin tagged fusion protein was used to colocalizated Vigilin with mRNA-decapping enzyme 1B (Dcp1B)which is a core member of the mRNA decapping complex and mRNA decapping complex initiated mRNA decay, indicate Vigilin is related to mRNA decay process.
