冬枣乙醛脱氢酶基因的克隆 与植物表达载体构建

摘要:醛脱氢酶( Aldehyde dehydrogenase, ALDH )可以将醛类物质转化为相应的羧基酸,是生物体内清除活性氧物质的重要酶类,对于维持生物机体的物质平衡有着重要作用。以冬枣半红期果实分离的总RNA为模板,经RT-PCR扩增得到1095bp的ALDH全长序列,与GenBank登录号为 JN831451.1的冬枣乙醛脱氢酶基因完全一致。此质粒经XbaⅠ和XmaⅠ限制性内切酶双酶切消化后,插入植物表达载体pBI121的35S启动子和NOS终止子之间,成功构建pBI121-ALDH正义表达载体。

Cloning and plant expression vector construction of aldehyde dehydrogenase gene in Ziziphus jujuba
Abstract:Aldehyde dehydrogenase (ALDH) can convert aldehydes into the corresponding carboxylic acid, scavenging the reactive oxygen species in vivo, which is important for maintaining the material balance of living organisms. Using the total RNA of half-red jujube fruit as template, the full-length sequence of ALDH was isolated by RT-PCR amplification, which is exactly the same as the aldehyde dehydrogenas gene of GenBank accession number JN831451.1. After digested by XbaⅠand XmaⅠrestriction enzyme, this plasmid was inserted between 35S promoter and NOS terminator of the expression vector pBI121 successfully.
