神经干细胞在表面改性PHBHHx膜上的存活及分化研究

摘要:目的 对PHBHHx膜进行表面改性,以增强与神经干细胞(NSCs)的组织相容性,进而为以NSCs为基础的脑组织工程应用奠定基础。方法 溶剂挥发法制备PHBHHx二维膜,扫描电镜观察其表面性状;分别通过脂肪酶处理,NaOH处理的方法对PHBHHx膜进行表面改性,测量接触角以检测膜表面亲水性;分离孕14.5天大鼠胚胎大脑皮质NSCs,培养至第三代的NSCs分别接种在表面改性后的PHBHHx膜表面,荧光染色观察细胞的分布,扫描电镜观察膜表面细胞形态,免疫细胞荧光化学观察NSCs分化情况。结果 与未处理的PHBHHx膜相比,脂肪酶、NaOH处理,接触角明显减小;NSCs在接种于PHBHHx支架后,能够继续存活,与未做表面改性处理组相比,处理后随着膜亲水性的增强,表面贴附细胞数量增加,存活状态良好,而且能够发生自发分化,分化为神经元和胶质细胞。结论 PHBHHx材料表面亲水性增强可以提高与NSCs的生物相容性。

The study of neural stem cells survival and differentiation on surface modified PHBHHx film
Abstract:Objective: In order to enhance the biocompatibility of PHBHHx films with neural stem cells (NSCs), we modified the surface of PHBHHx films, and then to lay the foundation for application of NSCs-based brain tissue engineering. Method: PHBHHx film was fabricated by a solution-casting method, and the morphology of the lm was observed under scanning electron microscopy(SEM); The film was treated by NaOH or lipase, then the surface hydrophilic property was characterized by water contact angle measurement; NSCs were isolated from the cerebral cortex of rat embryos on embryonic day 14-15, and P3 cells were seeded on surface treated PHBHHx films, after that the distribution of NSCs on films were studied by immunostaining, morphology of cells attach on the film were observed under SEM, Immune cell fluorescence chemistry was used to study the differentiation of NSCs. Result: Water contact angle of films treated with NaOH or lipase decreased dramatically compared with untreated; NSCs could survive on surface treated films, compare with untreated films the number of NSCs was increased, NSCs could also differentiate into neurons and glia cells on these films. Conclusion: The amelioration of hydrophilic property of PHBHHx film can improve its biocompatibility with NSCs.
