凝集素芯片对标记糖蛋白的定量

摘要:目的 进一步研究凝集素芯片对标记糖蛋白中定量的可行性. 方法 凝集素芯片是结合生物芯片技术和凝集素特性建立的检测糖蛋白糖链结构的糖组学新的重要研究工具。 结果 包被在基片上的凝集素与糖蛋白特异性亲和,荧光标记糖蛋白浓度与荧光信号值存在线性关系,1mg/ml是合适的凝集素点样浓度,凝集素芯片结合荧光探针技术非常敏感。结论 凝胶基片适合用来点制凝集素芯片,凝集素芯片可以精确定性和定量糖蛋白。

Lectin microarray used in the quantification of labeled glycoproteins
Abstract:Objective Further study of the feasibility of lectin microarray 's application in the quantification of labeled glycoproteins. Methods Integrated with the characteristics of lectin and fluorescence technique. Results Lectin microarray allows sensitive observation of multiple lectin-glycan interactions. 1mg/ml maybe the suitable lectin spotting concentration. It is feasible to analyze glycans structure of glycoproteins with the lectin microarray. Gel-slide is the suitable choice for the fabrication of lectin microarray. There is a linear relationship between the glycoprotein concentration and the fluorescence signal value. The lectin microarray is very sensitive. Conclution Lectin microarray can be used in the quantification of labeled glycoproteins.
