叶酸修饰PNiPAAm-co-PCl及其自组装纳米胶束的制备及表征

摘要:叶酸受体在多种肿瘤细胞中过表达是癌症治疗中一种可能的靶向药物结合位点。通过叶酸活性酯和N-羟乙基丙烯酰胺反应,制得叶酸修饰的N-羟乙基丙烯酰胺。核磁光谱显示叶酸成功连接在N-羟乙基丙烯酰胺上。通过原子转移自由基聚合合成叶酸修饰PNiPAAm-co-PCl,并通过超声乳化法制备了叶酸修饰的PNiPAAm-co-PCl自组装纳米胶束。考察胶束的形态、大小和温度敏感性。该方法可成功制备具有温度敏感性的叶酸修饰PNiPAAm-co-PCl纳米胶束,粒径约为100nm。

Preparation and Characterization of Folate-mediated PNiPAAm-co-PCl and its Self-assembled Nanomicelles
Abstract:Folic acid receptor was usually overexpressed in many cancer cells and was considered as potential target for targeted therapy agent. Folate-mediated N-hydroxyethyl acrylamide was prepared by the reaction of the activated folate ester and N-hydroxyethyl acrylamide. Nuclear magnetic resonance test that the folate was conjugated successfully with HAM. Folate-mediated PNiPAAm-co-PCl were formulated by ATRP. Self-assembled nanomicelles of folic acid grafted PNiPAAm-co-PCl were prepared by probe sonication method. The morphology and temperature sensitivity of folate-mediated PNiPAAm-co-PCl self-assembled nanomicelles were analyzed. Folate-mediated PNiPAAm-co-PCl temperature sensitive nanomicelles could be prepared successfully with the method. The average diameter of nanomicelles was about 100 nm.
