摘要:【目的】研究日粮不同硒水平对山羊后代Sepp1 和 apoER2 表达的影响。【方法】试验母羊共分为四组,分别为对照组(缺硒组),0.5 mg/kg(适量组)、2 mg/kg(高硒组)和4 mg/kg组(过量组)。提取山羊睾丸组织总RNA,应用实时荧光定量PCR方法检测睾丸组织中Sepp1 和 apoER2 mRNA在各组表达规律,利用免疫组织化学技术对睾丸Sepp1 和 apoER2进行定位研究和蛋白表达变化规律分析。【结果】0.5 mg/kg与2 mg/kg组Sepp1 and apoER2 mRNA及其蛋白的表达量显著高于对照组和4 mg/kg组(P<0.05),而对照组和4 mg/kg组之间没有显著差异。免疫组化定位及半定量分析得出与real-time RT-PCR 相似趋势的结果。【结论】不同日粮硒水平对山羊后代Sepp1 and apoER2的表达有影响,Sepp1 and apoER2有着相似的变化趋势,说明在生精上皮硒的吸收方面受体和配体的关系,apoer2是Sepp1的受体。

Effects of maternal dietary selenium (Se-enriched yeast) on the expression of Sepp1 and apoER2 of their offspring in goats
Abstract:In this experiment the effect of maternal dietary selenium on the expression of Sepp1 and apoER2 of their offspring in goats was studied. The experiment was conducted on 119 Taihang Black Goats randomly divided into 4 groups which were fed with a basal diet, were supplemented with 0 (control), 0.5, 2 and 4 mg kg1 DM Se. Testis samples were collected from the young male goats of each treatment group at the end of the study (30 d after weaning) for mRNA expression using real-time PCR and for protein expression by immunohistochemistry assay. Significant decrease was observed in mRNA expression of Sepp1 and apoER2 in the testis of Se-deficient (Group 1) and Se-excess (Group 4) compared with that in Groups 2 and 3. And a similar trend of the protein expression of Sepp1 and apoER2 was found. These data indicate that maternal and dietary selenium had an effect on the expression of Sepp1 and apoER2 in testis of their offspring. In addition, both gene had similar trend, which demonstrated that the relationship between Sepp1 and apoER2, and apoER2 is a receptor of Sepp1 in the seminiferous epithelium to uptake the selenium.
