AtSDG8 启动子解剖的初步研究

摘要:启动子控制着基因表达的起始和表达程度,对基因的活动起着"开关"作用。为了深入了解基因SDG8的功能与特性,我们通过探讨SDG8启动子的功能区域及基序组成,以GUS作为报告基因,选取了长度不同的两个启动子片段构建载体,转化得到两种拟南芥转基因植株SLP和SSP。对启动子的基序进行生物信息学分析后,选取了几种相关的外源激素ABA, MeJA和MeSA处理SLP和SSP植株。通过GUS基因的表达来推测SDG8启动子的功能区域以及基因对不同激素的应答反应。为进一步研究SDG8基因的结构与功能打下基础。

Preliminary Dissection Analysis of AtSDG8 Gene Promoter
Abstract:Promoter determines the initiation and level of gene expression and plays a role as a " switch" of gene activity. In order to have further understanding on the functions and characteristics of SDG8 gene, we selected two different fragments of SDG8 promoter to constructed vectors containing report gene GUS based on the analysis of the functional components and motifs of SDG8 promoter. We performed transformation on Arabidopsis Col-0 and obtained transgenic plants of both SLP and SSP We based on SDG8 promoter function area and characteristic, with GUS as a reporter gene, selects the length of two different promoter fragment .Based on bioinformatics analysis of the promoter motifs, we treated SLP and SSP plants with some relative hormones (ABA, MeJA and MeSA) respectively. We detected the response to different hormones of SDG8 promoter respectively and speculated the potential components of SDG8 promoter. Hence, this research may facilitate further research on SDG8 gene structure and functions.
