白藜芦醇对肥胖小鼠氧化还原状态和血压的影响

摘要:目的 研究白藜芦醇(resveratrol,RSV)对高脂性肥胖小鼠抗氧化能力和血压调节的作用及可能机制。方法 将4周龄雄性昆明种小鼠随机分成正常膳食组和高脂膳食(high fat die,HFD)组。到第12周时从高脂膳食组中选出高体重小鼠构成肥胖(Obesity)模型,再分成Obesity、Obesity+0.03%RSV、Obesity+0.06%RSV三个膳食组,与正常组一起继续喂养17周。结果 与肥胖组相比,0.06%RSV干预的小鼠体重显著降低,心组织抗氧化能力明显增强,炎症因子TNF-α、IL-6和MCP-1的含量显著降低(P﹤0.05),血管收缩活性因子血管紧张素Ⅱ和内皮素的含量显著降低(P﹤0.05),显著降低平均动脉压(MBP)(P﹤0.05)至接近正常组水平。其中氧自由基(ROS)、总抗氧化能力(T-AOC)、过氧化氢酶(CAT)、丙二醛(MDA)、血管紧张素Ⅱ(AngⅡ)都与MBP存在相关关系。结论 添加适量白藜芦醇可以有效抵抗高脂性肥胖小鼠的血压升高,其机制可能与抗氧化、抗炎症反应和调节血管活性物质含量有关。

Effects of Resveratrol on Redox State and Blood Pressure Regulation in Obese Mice
Abstract:Purpose To study effects and mechanisms of resveratrol(RSV) in high-fatted mice. Methods 4-week-old mice were divided into normal diet group as control and high-fat diet(HFD) group at random. In the 12th week, high body weight mice were choosed from high-fat diet group and formed obesity model. They were divided into 3 diet groups such as Obesity, Obesity+0.03%RSV and Obesity+0.06%RSV, and were fed another 17 weeks together with control. Results: Compared with Obesity, 0.06%RSV treatment markedly depressed body weight, obviously depressed antioxidant obilities of cardiac tissue, and also significantly reduced(P﹤0.05)contents of inflammatory factors like TNF-α, IL-6, MCP-1 and vasoactive factors like AngⅡ, ET in cardiac tissue. 0.06%RSV treatment significantly reduced(P﹤0.05)mean arterial pressure(MBP) to approach the lever of control group. RSV had doseeffect relationship.Therein, ROS, T-AOC, CAT, MDA, AngⅡall had correlativity with MBP. Conglutions Adding proper dose RSV could effectively resist blood pressure rise in high-fatted mice, and the machanisms might involve with its antioxidantive ability, ant-inflammatory reactions and regution of vasoactive matters contents.
