转化生长因子βⅡ受体在新生小鼠卵巢发育中表达

摘要:目的:探讨TβRⅡ在新生小鼠卵泡早期发育过程中的表达。方法:选择不同发育时期的C57小鼠卵巢,用免疫组化SP染色法,实时荧光定量聚合酶链反应(real time PCR)检测不同发育期卵巢组织中TβRⅡ蛋白、mRNA的表达水平。结果:TβRⅡ蛋白在各年龄段小鼠卵巢中的卵母细胞胞浆均有表达,在原始生殖细胞囊(germ cell cysts)中表达最强,始基卵泡中表达下降,在初级、次级卵泡中表达增强。初级卵泡的颗粒细胞开始表达TβRⅡ,并随着卵泡发育表达逐渐增强。TβRⅡ mRNA在卵巢组织中的表达水平随卵巢的生长发育呈先降低后增高的趋势。0、1、11天小鼠卵巢中TβRⅡ mRNA表达水平无统计学差异(p > 0.05),但其表达水平是2、5、7天小鼠卵巢组织的两倍(p < 0.05)。结论:TβRⅡ在卵泡发育初始阶段的表达具有时间和空间的特异性,提示TGFβ信号通路在早期卵泡发育过程中起着重要的调节作用。

Expression of Transforming Growth Factors-βⅡReceptor during Early Folliculogenesis of neonatal mice
Abstract:Objective: To investigate the expressions of protein and mRNA of TβRⅡ during early folliculogenesis of the neonatal mice ovary. Method: Ovaries were collected from the neonatal mice of different age stages. The expressions of TβRⅡ proteins and mRNA were detected and evaluated by immunohistochemistry and real-time polymerase chain reaction respectively. Results: TβRⅡ proteins were expressed in the cytoplasm of oocytes within ovaries in different age stages. The highest intensity of TβRⅡ staining was present in germ cell cysts. Primordial follicles exhibited moderate staining, and the staining of TβRⅡ increased during the transition to in primary and secondary follicles. For granulosa cells, TβRⅡ expression was found to be primarily in the primary follicles and increased with follicular development. There was no significant difference in TβRⅡ mRNA level among the ovary tissues of 0 day, 1 day, 11 days (p > 0.05). However, the ovarian TβRⅡ mRNA level during these periods was 2-fold higher than these of 2 days, 5 days and 7 days(p < 0.05). Conclusion: The expression of TβRⅡ proteins and mRNA in neonatal mice ovary suggests that the TGFβ signaling pathway may play an important role during early folliculogenesis.
