重组酵母的淀粉乙醇直接发酵法工艺条件优化

摘要:本研究对α-淀粉酶和糖化酶双基因重组酵母工程菌的乙醇发酵条件进行探讨,研究了该菌株在糖蜜种子培养基中的出芽率、死亡率以及生长动力学,并通过正交试验对淀粉发酵培养基成分以及发酵条件进行了优化,得出其最佳直接发酵条件为:将一级酵母种子接入含初始淀粉150 g/l的最优发酵培养基中,接种量10%,发酵6d后得到发酵液中乙醇体积分数为8.3%,乙醇转化率达到理论值的78.1%。按照本文确定的乙醇直接发酵法工艺,利用具有α-淀粉酶和糖化酶分泌能力的工程酵母对淀粉直接发酵生产乙醇,取得良好的效果,具有重要的经济价值和社会意义。

Optimization of Direct Ethanol Fermentation Conditions from Starch with Recombinant Yeast
Abstract:In this paper, the direct ethanol fermentation process from starch using a recombinant strain of Saccharomyces cerevisiae co-expressing alpha-amylase and glucoamylase was studied. The budding rate, mortality rate and growth kinetics of yeast in molasses medium were investigated. Fermentation medium composition and process conditions were also optimized through Orthogonal Experiments. When the first order seed was inoculated to optimal fermentation medium containing 150 g/l of soluble starch by 10%, 8.3% (v/v) of ethanol was received after 6 days, approximate to 78.1% of the theoretical yield. It can be concluded that the direct fermenting process using our recombinant yeast established in this research got positive results and showed important economic value.
