柽柳ThHSP1基因在酵母中的抗逆性分析

摘要:热激蛋白(Heat shock proteins, HSPs)是生物体受热或其他环境胁迫后诱导表达的一类应激蛋白。本研究用酵母表达系统研究了柽柳HSP基因(命名为ThHSP1)的抗逆性。将ThHSP1基因构建到酵母表达载体pYES2中,转化到酿酒酵母INVSc1菌株中,并以转空pYES2质粒的酵母INVSc1(pYES2)作对照,比较两种酵母在干旱(Sorbitol)、重金属(CuSO4和ZnCl2)、盐(NaCl、KCl、MgCl2、NaHCO3)、低温(-20℃)和高温(53℃)胁迫下的存活率,结果表明,ThHSP1基因提高了转基因酵母的抗各种胁迫的能力。

Stress tolerance analysis of ThHSP1 gene from Tamarix hispida in yeast
Abstract:Heat shock protein (Heat shock proteins, HSPs) are a kind of stress protein that induced by heat or other environmental stress in organism. In the present study, the recombinant yeast expression system was used study the stress tolerance of ThHSP from Tamarix hispida. The ThHSP1 was cloned into the pYES2 vector and transformed into yeast Saccharomyces cerevisiae INVSc1. And the yeast INVSc1 transformed with an empty pYES2 vector was employed as a control. The survival rates between the recombinant yeast INVSc1 (pYES2-ThHSP1) and control INVSc1 harboring empty pYES2 were compared under drought (Sorbitol),heavy metals(CuSO4 and ZnCl2),Salts(NaCl、KCl、MgCl2 and NaHCO3),freezing(-20℃)and heat(53℃)stresses. These results demonstrated that ThHSP1 enhanced the tolerance to a variety of stress conditions in yeast.
