荧光定量PCR方法对茶树花蕾不同发育时期CsPSP1基因表达量的研究

摘要:本实验以黄山早芽、乌牛早、舒茶早、龙井43四个茶树品种的花蕾为材料,并将每个品种的花蕾按发育的早、中、晚三个期进行分类。以早期花蕾为对照组比较花蕾发育的各个时期基因表达量的差异,利用荧光定量PCR的方法确定不同时期花蕾的CsPSP基因表达量,。实验结果表明, 黄山早芽、乌牛早、舒茶早、龙井 43四个品种的CsPSP基因表达量,中期花蕾分别为早期花蕾的1.8、2.7、2.5、6.5倍;晚期花蕾分别为早期花蕾的2.5、3.7、3.1、44.2倍。以上结果表明在发育过程中,CsPSP.基因的表达量.呈上升趋势,其中龙井43花蕾的早、中、晚三个阶段的基因表达量增加显著,这可能由品种的差异造成的。通过对实验数据的分析,推断CsPSP基因为茶花发育的晚期基因。

Research of CsPSP1 gene expression amount of different development stages in Tea buds by real-time Fluorescence quantitative PCR
Abstract:This experiment taken use of the four varieties in tea tree buds as material, huangshanzaoya, wuniuzao, shuchazao, longjing 43. And each species of bud was classified as the early, middle and late stage according to size. Taking early bud as reference, the CsPSP gene expression was determined by real-time fluorescence quantitative PCR method about different periods of buds. The results show that middle period bud CsPSP gene expression of huangshanzaoya, wuniuzao, shuchazao, longjing 43 were 1.8, 2.7, 2.5, 6.5 times of early period bud respectively; late period were 2.5, 3.7, 3.1, 44.2 times of early period bud respectively. This Suggests that CsPSP gene expression has been on the rise in the development process in each variety bud . The differences of gene expression is very conspicuous in longjing 43 three development stages. It might be associated with varieties. Through analysing the experiment data, it could be deduced that the CsPSP gene is the late gene in the development process of tea flower.
