HPLC同时检测连翘酯苷、连翘苷和连翘脂素方法的建立

摘要:目的 建立简单有效的同时测定连翘酯苷A、连翘苷和连翘脂素的Hplc法。方法 以甲醇和0.1%甲酸水为流动相,流速为1.0 mLomin-1,检测波长为227nm、332nm,柱温30℃,进样量20ul,经Hypersil ODS C18柱 (250 mm×4.6 mm, 5μ m)分离,进行色谱分析。结果 本方法连翘酯苷A的r=0.9999,线性范围为3.65~116.67μg;连翘苷的r=0.9999,线性范围为0.31~10μg;连翘脂素的r=0.9999,线性范围为0.31~9.89μg,平均回收率(n=5)分别为99.59%、99.4%和99%,RSD为0.56%、 0.96%和 0.91%。精密度和重复性试验RSD均小于5%。结论 本方法简单、灵敏,重现性好,可用于同时测定连翘药材中的连翘酯苷、连翘苷和连翘脂素。

Determination of forsythoside A, phillyrin and phillygenin by HPLC
Abstract:Objective To establish a method for the determination of forsythoside A , phillyrin and phillygenin by HPLC. Method Methanol-water-foemic acid was taken as the mobile phase at a flow rate of 1.0 m L/min. The detective wavelength was 277 nm and 332nm.The temperature was 25℃ and the sample volume was 20uL.The Hypersil ODS C18 column (250 mm×4.6 mm, 5μ m) was taken as column for the separation and then carried out the chromatographic analysis. Result The linear rangs of forsythoside A , phillyrin and phillygenin were 3.65~116.67μg, r=0.9999 and 0.31~10μg , r=0.9999 and 0.31~9.89μg, r= 0.9999.The average recoveries(n=5) were 99.59% (RSD=0.56%),99.4% (RSD=0.96%) and 99% (RSD=0.91%). RSD of precision and repeatability test was less than five percent. Conclusion The results show that the method is simple , sensitive and highly reproductive , and it is suitable for the determination of forsythoside A , phillyrin and phillygenin.
