离子交换树脂固定接枝脂肪酶的优化研究

摘要:采用吸附法和吸附-交联法将碱性脂肪酶L4固定接枝在阳离子交换树脂DK110上。考察了酶浓度、温度、pH值等因素对酶固定化的影响,并在不同温度和pH条件下,考察了固定化酶与游离酶的性能对比。结果表明:酶液浓度、温度和pH值对脂肪酶固定化影响显著,最佳固定化条件为:酶液浓度为10-20mg/mL、温度为50℃、酶液pH值为5。所得固定酶耐受温度和pH变化的能力增强,而且吸附-交联法获得的固定酶稳定性更好。

Optimization of Lipase Immobilization on Ion Exchange Resin by Adsorption
Abstract:L4 Lipase was immobilized on ion exchange resin (DK110) by two methods namely adsorption and adsorption followed by cross-linking. The influences of lipase concentration, adsorption temperature, pH value on the adsorption and activity of immobilized lipase were investigated in this paper. The results show that: lipase concentration, temperature, pH value have great impact on the immobilization of lipase. The optimal conditions for immobilization: the amount of lipase 10-20mg/mL, adsorption temperature 50℃, buffer pH value 5. Furthermore, immobilized lipases are much more stable than free lipase at different temperatures and pH. Lipases immobilized by adsorption followed by glutaraldehyde treatment exhibited higher activities and stabilities.
