基于金胶的表面增强拉曼光谱(SERS)检测食源性致病菌

摘要:食源性致病菌是威胁食品安全的重要因素之一,其中肠道致病菌,无芽孢,周身鞭毛或者无鞭毛,是爆发率较高的一大类,由其引发的食品污染不仅危害人的身体健康,而且不利于食品贸易。本文研究应用表面增强拉曼光谱(SERS)技术检测四个种属共7种致病菌:包括痢疾志贺氏菌、鲍氏志贺氏菌、宋内氏志贺氏菌在内的志贺氏菌属;包括鼠伤寒沙门氏菌、沙门氏菌O7HZ10在内的沙门氏菌属;柠檬酸杆菌属的弗氏柠檬酸杆菌和肠杆菌属的阪崎肠杆菌。同时,分别针对不同批次的金胶和阪崎肠杆菌进行SERS增强效果的重复性和稳定性验证。研究了高压蒸汽灭菌对致病菌SERS图谱的影响,对7种致病菌进行主成分分析和系统聚类分析。研究表明,在SERS稳定性较好的情况下,7种菌的SERS图谱比对文献并进行特定峰的归属,应用主成分分析和系统聚类分析进行分类后能较明显的区分各属和各个菌种,建立了对多种食源性致病菌的快速定性鉴别。

Surface enhanced Raman spectroscopy (SERS) to detect foodborne pathogens by gold nanoparticles
Abstract:Forborne pathogens are very important threats to food safety, in which intestinal pathogens, nonspore-bearing , the whole body flagella or flagella , is a large class of high outbreaks of food contamination caused by not only hazardous to human health, but also not conducive to the food trade. Therefore, this paper studies the application of gold nanoparticle surface enhanced Raman spectroscopy (SERS) on detection four kinds of pathogenic bacteria: Shigella- Shigella dysenteriae, Shigella boydii,Shigella sonnei ; Salmonella - Salmonella typhimurium, Salmonella O7HZ10; Citrobacter - Citrobacter freundii and Enterobacter - Enterobacter sakazakii. At the same time, different batches of gold colloid and Enterobacter sakazakii repetitive verification SERS enhancement and stability. Second, it studied the impact of the SERS spectra of high pressure steam sterilization of pathogenic. Finally, seven kinds of pathogens by principal component analysis (Principal Component Analysis, PCA) and cluster analysis (Hierarchical Cluster Analysis HCA). Studies have shown that the SERS spectra of the seven kinds of bacteria than the literature and the attribution of a particular peak in the case of SERS good stability, the application of PCA and HCA to classify the more obvious distinction between each genus and each species. At the same time, before and after the autoclave sterilization SERS can be seen that the bacterial protein occurred degeneration.
