芸薹种SSR和UGMS标记遗传连锁图谱的构建

摘要:本研究以芸薹种大白菜亚种多国芸薹属AA基因组测序计划(The Multinational B rassica Genome Project, MBGP)模式植物'Chiifu'为父本,以小白菜亚种高代自交系'四九菜心'为母本,利用国际公用简单重复序列(Simple sequence repeats, SSR)标记和本实验室设计开发的单基因微卫星(Unigene microsatelite sequence, UGMS)标记,以BC1代为作图群体,构建了一张总长度为1160 cM,包含有10条连锁群的分子遗传图谱。本图谱共计251个分子标记,包含120个UGMS和131个SSR标记。单拷贝标记数位218个。标记的平均图距为4.62 cM。每条连锁群有15至37个标记,连锁群长度介于79.3 cM-150.9 cM之间。该图谱为构建一张完整的大白菜染色体片段替换系奠定了基础。

The genetic linkage map of Brassica rapa based on SSR and UGMS markers
Abstract:A BC1 mapping population derived from a cross between two subspecies of Brassica rapa, the model plant material of the Multinational Brassica Genome Project 'Chiifu' (B. rapa spp pekinensis) as donor parent and an inbred line 'caixin' (B. rapa L. ssp. chinensis) as recurrent parent, was used to construct a linkage map of B. rapa. Linkage analysis of 251 polymorphic markers, including 131 public SSR and 120 UGMS markers, gave 10 linkage groups (LGs) with a total genome coverage of 1160 cM. The average distance between adjacent loci was 4.62 cM. The number of markers on each LG was ranged from 15 to 37, and the length of LG from 79.3 cM to 150.9 cM. This map will lay a foundation for construction of chromosome segments substitution lines of Chinese cabbage.
