款冬花提取物抗氧化活性及对小肠α-葡萄糖苷酶抑制作用的研究

摘要:研究款冬花提取物(CET)中多酚类物质的抗氧化活性,并用Caco-2细胞模型分析CET对小肠α-葡萄糖苷酶活性的抑制作用。实验测得CET中总酚与总黄酮含量分别为116 mg没食子酸当量/ g提取物与181 mg芦丁当量/ g提取物。经高效液相色谱分析,得到绿原酸(38.72 mg/g)、3,4-二咖啡酰奎尼酸(20.64 mg/g)、3,5-二咖啡酰奎尼酸(15.77 mg/g)、4,5-二咖啡酰奎尼酸(9.69 mg/g)及芦丁(119.36 mg/g)为CET中主要的活性物质。CET清除ABTS、DPPH自由基的半数有效浓度(EC50)分别为1.254 mg/mL、0.055 mg/mL。将提取物(2.5 mg/mL)作用于经H2O2 (2 mM)处理过的Caco-2细胞,细胞活力提高40.49 %。CET (2.5 mg/mL)作用于Caco-2细胞单层模型,对细胞AP侧和BL侧麦芽糖酶活性的抑制率分别为56.84 %和56.74 %。以上结果表明,款冬花可作为一种潜在的资源应用于抗氧化和抗餐后高血糖保健食品的开发。

Antioxidant and α-glucosidase Inhibitory Activities of Tussilago Farfara L. flower buds
Abstract:This study was performed to investigate antioxidant and a-glucocidase inhibitory activities of the crude extract from flower buds of Tussilago farfara L. (CET) together with detecting the main active compounds by HPLC. The total phenols and flavonoids contents were 116 mg gallic acid equivalent/g and 181 mg rutin equivalent/g, respectively. Chlorogenic acid (38.72 mg/g), 3,4-dicafeoylquinic acid (20.64 mg/g), 3,5-dicafeoylquinic acid (15.77 mg/g), 4,5-dicafeoylquinic acid (9.69 mg/g) and rutin (119.36 mg/g) were detected as the main active compounds of CET by HPLC analysis. CET exhibited a potent scavenging effect on ABTS and DPPH free radicals with EC50 values of 1.254 mg/mL and 0.055 mg/mL, respectively. CET (2.5 mg/mL) exhibited a protecting effect on Caco-2 cells when treated with 2 mM H2O2. This extract showed an inhibitory effect against the maltase enzyme with 56.74% inhibition on apical side and 56.84% inhibition on basal side of Caco-2 monolayer at a concentration of 2.5 mg/mL. The result suggested that Tussilago farfara could be used as a potential antidiabetic and antioxidation material in food industry.
