季铵盐对巨型囊泡形成的影响

摘要:本文用季铵盐与蛋黄卵磷脂混合,利用电形成法构建了10-100μm的巨型单层囊泡,并将碳纤维超微电极应用于电形成法制备囊泡的研究中。通过在碳纤维电极间施加电位峰峰值Epp = 0.212-20V/mm,频率f = 1-20MHz的交流电场,分别在水溶液与缓冲溶液中调控囊泡的形成。并通过倒置光学显微镜实时原位考察了交流电场、季铵盐疏水链链长以及缓冲溶液对囊泡形成的影响。研究发现在20Hz 恒定频率下,囊泡的形成速率随着电压的增加而加快。而且囊泡的平均直径随着掺入的季铵盐疏水链链长的增加而增加。此外,在实验中,缓冲溶液的加入会降低囊泡尺寸,延长囊泡的存在时间。

Influence of Quaternary Ammonium Salts on the Formation of Catanionic Vesicles
Abstract:In this paper, the electroformation method was used in constructing giant unilamellar vesicles(GUVs) with the mixture of quaternary ammonium salt and EggPC, and carbon fiber was used as ultramicroelectrode in this progress. Giant vesicles were obtained over a wide frequency range of 1-20MHz and peak-to-peak value from 0.212V/mm to 20V/mm between two parallel conducting electrodes of carbon fibers under alternative current (AC) electric field in distilled water or buffer solution. The influence of AC electric field, chain length of quaternary ammonium salt and buffer solution on the structural evolution of giant vesicles was in situ observed by inverted optical microscope. Our study demonstrates that the formation time of vesicles decreases with the voltage increases and the average diameter of the catanionic vesicles increases with the chain length of quaternary ammonium salts increases. Moreover, it is found that the application of buffer solution diminish the size of vesicles and extend the existence time of catanionics vesicles.
