亲和素修饰的硅壳荧光纳米探针用于肺癌细胞的检测

摘要:将荧光纳米标记技术与免疫荧光分析法相结合,发展了一种新型的荧光纳米探针间接荧光显微镜法用于肿瘤细胞中癌胚抗原的检测。先用兔抗癌胚抗原抗体特异性地识别人肺癌细胞中的癌胚抗原蛋白,再通过生物素化羊抗兔抗体与兔抗癌胚抗原抗体特异性结合,再以修饰亲和素的包被异硫氰酸罗丹明荧光染料的二氧化硅荧光纳米粒子作为信号指示剂,利用亲和素与生物素的特异性结合,实现对肺癌细胞中癌胚抗原的特异性标记,最后通过荧光显微镜对标记的肿瘤细胞进行荧光成像检测。结果表明该探针能够有效地识别肺癌细胞表面的肿瘤标志物-癌胚抗原。

Detection of lung carcinoma cells using avidin-conjugated dye-doped silica nanoprobes
Abstract:A method of fluorescent nanoparticle-based indirect immunofluorescnce microscopy has been developed for the rapid detection of carcinoembryonic antigen (CEA) in human lung carcinoma A549 cells. A rabbit anti-CEA IgG was used as primary antibody to recongnize CEA in human lung carcinoma A549 cells, a specific biotinylated goat anti-rabbit IgG was used to recongnize the rabbit IgG, and then avidin-labeled tetramethylrhodamine-5-isothiocyanate (TRITC)-doped silica nanoparticles was used to generate fluorescent singal for microscopic examination. The result shows that the nanoprobe can be effectively applied for the recognition of CEA protein in human lung carcinoma A549 cells.
