百合多糖体外免疫调节活性研究

摘要:采用体外巨噬细胞RAW264.7模型研究百合多糖体外免疫调节活性,结果表明,百合多糖LLPS-1和LLPS-3对小鼠巨噬细胞增殖活性影响是随着浓度增大先升高后降低,100 μg/mL达到最大值;LLPS-2则是增殖活性一直升高,在400 μg/mL时和阳性对照LPS无显著差异。3种百合多糖组分随浓度增大,刺激巨噬细胞吞噬中性红的能力先增强后略微减弱,在50-400 μg/mL浓度范围内显著高于空白对照。3种组分对NO释放的影响是随着浓度增大,释放的NO浓度逐渐升高,其活性呈一定的剂量的依赖性,在很低浓度(25 μg/mL)就能极显著促进巨噬细胞释放NO,在400 μg/mL时显著高于空白对照和阳性对照。总的来说,体外免疫调节活性强弱的顺序是:LLPS-2﹥LLPS-3﹥LLPS-1。

Research on immunomodulating activities in vitro of LLPS
Abstract:Immunomodulating activities in vitro of LLPS were measured by using macrophage RAW264.7 model experiments. Results show that, proliferation of macrophage increased and then decreased with the increasing concentration of LLPS-1 and LLPS-3, and reached their maximum at the concentration of 100 μg/ mL. The effect of LLPS-2 on proliferation of macrophage had been increasing, and there was no significant difference between LLPS-2 and the positive control LPS at the concentration of 100 μg/ mL. The ability of LLPS to stimulate macrophages to devour neutral red was enhanced at first and then slightly weakened with the increasing concentration. In the range concentration of 50-400 μg/mL, LLPS were significantly higher than that of the blank control. LLPS could promote NO production of macrophages and their activities were dose-dependent. They would be able to significantly promote macrophages release NO at low concentration (25 μg/mL) and NO production significantly higher than the blank control and positive control at the concentration of 400 μg/mL. In general, the order of immunomodulating activity in vitro is LLPS-2> LLPS-3> LLPS-1.
